ULTIMATES と Spider-Man のクロスオーバーです。
タイトルの"SIX"は6人で、オズボーン（グリーンゴブリン）のいうところの、
Green Goblin = Norman Osborn
Doctor Octopus = Otto Octavius
Electro = Max Dillon
Sandman = Flint Marko
Kraven The Hunter = Sergei Kravinoff
Spider-Man = Peter Parker
というメンバーになります。
ピーターは、メイおばさんを盾に脅されて、ほんの一瞬だけ仲間になりますが、実質5人で行動しています。

オズボーンは薬なしでグリーンゴブリンに変化することができるようになり、ドクターオクトパスはアームを遠隔操作できるようになったりと進化を遂げています。
ハンターも、以前は普通の人間だったのですが、遺伝子操作をしてパワーアップしていますが、相変わらず雑魚扱いです。

ULTIMATES のメンバーとしては、ハンク（ジャイアントマン）とジャネット（ワスプ）がまだ夫婦でいるあたりなどを考えると、時期的に「１」の真ん中あたりといったところでしょうか。

大風呂敷を広げた割には、結構終りはあっけなく、戦いの見せ場が物足りないような感じがしました。
あと、今回の人物の顔の描き方が何か嫌で、ピーターの顔なんてかなり哀れでした。
でもまあ、最後のキャプテン・アメリカの台詞

It's men of influence and power that decide what these wars will be about.
They decide who we are going to fight and how we will fight them...
And then they go about planning the fight.
In a sense, really, people of power will the war into existence.
